Carb's too big. To use that size of a carb you need one with an accelerator pump. Even then, on a street engine, it's too big to snap the throttle open and not expect it to bog until the RPM is up in probably the 3500+ range or more. It's not a workable carb on a stock engine.

That was written when the OP mistakenly wrote he'd got a 42mm carb fitted, whereas it is actually a 34mm.
 
My 850 has a single Mikuni. I don't let it run with the choke on at all. Fuel on, choke on (ignition off), kick through 3 times, choke off, ignition on and start. You have to stay on the throttle for a bit until it will idle. I had the same problem fouling plugs using the choke.
 
Likewise, I have 850 with single Mikuni. Fuel on, enrichment circuit on (choke), ignition on, hit starter button :), leave "choke"
on for 20-30 seconds then shut it off and work the throttle until it will idle. Have never had a fouled plug or any sign of
black sooty carbon on the plug.
 
In addition to the advice above with starting, try adding a slide spring from an amal carb. It will fit right inside the Mikuni spring and add a bit more "snap" to the throttle action, and keep you from piling it on too suddenly.
 
My bike ,with same setup never idles on choke as soon as it fires I flip choke off ( lever mounted on carb) then occasionally have to blip throttle depending on temperature , a short ride through town to good backroad can blacken plugs , so I avoid that type of ride , just hit the good roads and go , no victory laps down main drag type of thing , plugs stay nice for ages when rides are of this type , just puttin’ around town will blacken them quickly ....

As fast as you physically can?
Or it bogs trying to ride it?
I don’t know your experience or skill set, so a few Q’s.

Or falls flat?
 
Here's the latest up date..did a considerable amount of messing around with the timing and carburation and it starts and runs fine, throttle response is crisp and the plugs are burning perfectly. Low speed performance isn't that good though and I think it's a carb adjustment problem, maybe just a simple adjustment??

What throttle position isn't good, and what are the symptoms? 1/8th vs 1/4 vs 1/3 throttle are different circuits.
