Why can't I get my bike started without starting fluid when it has sat for a few days. Once started it's so easy to restart, even the next day. 75 Commando with Mikuni single. Carb has been rebuilt, bike is tuned up...WTF
 
First thing the occurs to me is float level a bit too low. Proper tune will require some enriching for first cold start, ticked or chocked or spritzed aerosol. Check for possible air leaks and of course the ever famous pilot jet cruded up.

Don't take offense, don't know how long you've had the Mik. New problem? Always been like that? Very important with a Mik to crank with the throttle COMPLETELY CLOSED and the enrichener (choke) on. Opening the throttle even the smallest amount kills the ability to draw fuel in.
 
I'll go with hobot, low float. Does it tickle good? If not, low float.

+1 Been there. One more reason to go with a good synthetic multi 8)

+2 Went from 50w to 30w. Starts easy now but this summer will be back to 50w.
 
VM Mikuni should have a starter jet at the bottom of the long tube like hole in the float bowl. Drill jet bigger till choke works. Another bike I got TM Mikuni's. It came with 15 starter jet. I ended with hole twice as big or 4 times volumne before it worked good.
